A confocal image showing microglial phagocytosis in AD mouse model (3xTg, 3-month-old). Image acquired with my students at the Bordeaux School of Neuroscience, 07/2024, Bordeaux, France. Note, green, IBA1; red, amyloid-beta.

Brain slide scannings
(Immunofluorescence)

A brief explanation on these images acquired by me previously:

  • Figure 1 (unpublished): overexpression of a protein (we identifed from dopamingergic neuron profiling), in a TH-Cre driver mouse line (unpublished).
  • Figure 2 (unpublished):  enlarged view in Figure 1.
  • Figure 3: examing dorsomedial nucleus accumbens (dmNAc) Tac2-expressing cells and their projection.
  • Figure 4 (unpublished): injecting a Cre-dependent Gq virus into dmNAc in Tac2-Cre mice.
  • Figure 5 (unpublished): Th+ cells in the midbrain, in a genetic mutant mice (related to diabetes project).
  • Figure 6: expression pattern of the dmPAG Tac2+ cells.
  • Figure 7 (unpublished). Using TMEM119-EGFP mice to examine how short-term HFD affects microglia morphology (brain slide). Image acquired together with my student Emily (summer student working with me).
  • Figure 8 (unpublished). Confocal image from a TMEM119-EGFP mouse fed with short-term HFD. We aimed to examine microglia morphology in a higher maganification. Image acquired together with my student Emily (summer student working with me).

Phagocytosis, Abeta Uptake
(Images or live-cell imaging)

Examing phagocytosis in astrocytes or microglia. To establish this important functional readout, I have to use multiple ways to perform this experiment. These images are also acquired by me (unless further noted)

  • Figure 1: iGIRKO/5xFAD mice, examing Abeta uptake using IHC (GFAP and Abeta).
  • Figure 2 (demo experiment): in 3xTg mice, examing microglia phagocytosis (IBA1 and Abeta). Image acquired by me and my proud students in Bordeaux School of Neuroscience (07/2024, Bordeaux, France)
  • Figure 3 (unpublished): live cell imaging, examined by Incucyte S3. SIIM-A9 cells uptaking Abeta555.
  • Figure 4 (unpublished): whole-brain CLARITY, examing blood vessel (lectin) and Abeta deposition, in 5xFAD mouse brain.
  • Figute 5 (demo experiment): in BV2 cells, examing cellular uptake of Abeta555. Image acquired by me and my proud students in Bordeaux School of Neuroscience (07/2024, Bordeaux, France)
  • Figure 6 (unpublished): examing gliosis (or reactive astrocyte) in 5xFAD brain (with or without insulin receptor KO)
  • Figure 7 and Figure 8 (unpublished): SIM-A9 cells uptaking E.coli(488). Cells are fixed but not stained.

Abeta deposition along blood vessels in the brain, is also one of my interested areas. Using whole-brain CLARITY, I am studying how astrocyte insulin receptor (IR) KO can affect vascular amyloid deposition.

  • Right: 5xFAD or iGIRKO/5xFAD mouse brain (sorry I will not tell you what it is until this is published)
